Target Information
ARC (also known as NOL3 - Nucleolar protein 3) has two isoforms.
- Isoform 1: involved in RNA splicing.
- Isoform 2: functions as an apoptosis repressor blocking multiple modes of cell death.
ARC inhibits extrinsic apoptotic pathways by:
- Interacting with FAS and FADD upon FAS activation, blocking DISC assembly.
- Interacting with CASP8 in a mitochondria localization- and phosphorylation-dependent manner, limiting soluble CASP8 available for DISC-mediated activation.
Additionally, ARC inhibits intrinsic apoptotic pathways by interacting with BAX, resulting in BAX inactivation and preventing mitochondrial dysfunction and release of pro-apoptotic factors.
Mutations in this gene can result in myoclonus, familial cortical.
