Product Specific Information

The 1B1 monoclonal antibody reacts with mouse CD1d, a 48 kDa glycoprotein structurally homologous to MHC class I molecules. CD1d associates with β2-microglobulin and functionally resembles MHC class II. The 1B1 clone detects CD1d at varying levels on mouse leukocytes and β2m-associated CD1d.

• Applications Tested: Tested on mouse splenocytes; ≤0.125 µg/test (100 µL volume, 10⁵–10⁸ cells/test).
  Antibody concentration should be titrated for optimal performance.

Super Bright 645:

• Tandem dye excited at 405 nm (violet laser) and emits at 645 nm.
• Recommended filter: 660/20 bandpass.
• Use Super Bright Complete Staining Buffer (SB-4401) when using multiple Super Bright dyes to minimize non-specific polymer interactions.

Light Sensitivity:
This dye is sensitive to photo-induced oxidation; protect vial and samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (00-8222)
• 1-step Fix/Lyse Solution (00-5333)

Target Information

CD1d is a heavy chain associated with β2-microglobulin on cortical thymocytes. It can also be expressed independently on human intestinal epithelial cells. CD1d is the sole group 2 member of the CD1 family of MHC-like glycoproteins. These proteins present lipid and glycolipid antigens to T cells. The CD1 family genes are located on chromosome 1 and differ in localization and ligand specificity. CD1d localizes to late endosomes and lysosomes via a tyrosine-based motif.
Diseases associated with CD1D include Mycobacterium Malmoense infection and autoimmune disorders.
