Product Specific Information
The SA-DA4 monoclonal antibody reacts with the mu heavy chain of human IgM and does not cross-react with other immunoglobulin classes (IgD, IgG, IgA). IgM is expressed intracellularly during early B cell development and on the surface of mature B cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test.
Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.
Staining Recommendations:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.
Fixation:
Samples may be stored up to 3 days at 2–8°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.
