Target Information
The PKC family of serine/threonine kinases, including PRKCG (PKC gamma), is activated intracellularly by signal transduction pathways. In humans, at least 12 different PKC polypeptides have been identified, differing in structure, distribution, localization, and substrate specificity. PKC alpha, beta I, beta II, and gamma form the conventional family, which are Ca²⁺- and phospholipid-dependent.
PKC family members phosphorylate diverse protein targets and play roles in various signaling pathways. They also act as receptors for phorbol esters, a class of tumor promoters. PKC gamma is expressed exclusively in the brain and spinal cord, localized to neurons, and is essential for neuronal functions such as long-term potentiation (LTP) and long-term depression (LTD). Knockout studies suggest involvement in neuropathic pain development. Mutations in PKC gamma have been linked to spinocerebellar ataxia-14 (SCA14).
