Target Information
Transglutaminase (TGase)는 단백질 간의 e-(γ-glutamyl) lysine isopeptide 결합을 형성하여 교차결합을 촉매하며, 활성에는 Ca²⁺ 결합이 필요합니다. 포유류에서는 8가지 TGase 동형효소가 확인되었으며, 그중 Tissue transglutaminase (tTGase, TGase 2)는 N-말단 β-sandwich, catalytic core, 두 개의 C-말단 β-barrel 도메인을 가지고 있습니다. tTGase는 세포사멸, 세포증식, 세포분화, 수용체 매개 세포내섭취 등에 관여할 수 있습니다. 알츠하이머병 뇌에서는 tTGase 활성 증가로 인해 Ab peptide, β-amyloid precursor protein, tau protein 등의 중합이 일어나며, 이는 신경섬유 엉킴 형성으로 이어집니다.
