Target Information
NFS1 (Nitrogen Fixation 1), also known as NIFS or IscS (cysteine desulfurase), belongs to the class V pyridoxal-phosphate-dependent aminotransferase family.
It localizes to the cytoplasm or mitochondrion depending on cytosolic pH. NFS1 shows highest expression in heart and skeletal muscle, with lower expression in liver, brain, and pancreas.
NFS1 catalyzes the removal of sulfur from cysteine to form alanine, providing inorganic sulfur for iron-sulfur (Fe-S) clusters. These Fe-S clusters are essential cofactors in various biological processes such as:
- Electron transfer in oxidative phosphorylation
- Enzymatic catalysis in aconitase and dehydratases
- Structural maintenance in DNA repair enzyme endonuclease III
