Description:
This OX33 monoclonal antibody reacts with rat CD45RA. In rats, this isoform of CD45 (leukocyte common antigen) is present only on B cells. CD45 is a type I transmembrane protein tyrosine phosphatase existing as multiple isoforms due to alternative splicing.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.25 µg per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Fluorophore Characteristics:
APC-eFluor 780 emits at 780 nm, excited with a red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, lymphocyte development, and antigen signaling.

CD45 isoforms differ in extracellular domains due to alternative splicing but share identical transmembrane and cytoplasmic domains. Expression varies by cell type and physiological state. CD45 also modulates integrin-mediated signaling and participates in apoptosis-related DNA fragmentation.

CD45 is expressed in human hematopoietic cells except mature red cells and is absent in non-hematopoietic tissues. Dysfunctions are associated with severe combined immunodeficiency and Hepatitis C.
