Target Information
Cyclin D1 (PRAD-1, bcl-1) is a key regulator of the cell cycle that functions with CDK4 and/or CDK6 to phosphorylate the Rb protein. It acts as a proto-oncogene, frequently overexpressed in various human cancers including mantle cell lymphoma. Cyclin D1 regulates protein phosphorylation, cell proliferation, and differentiation processes.
The CCND1 gene is located on chromosome 11 and encodes a cytoplasmic and nuclear protein synthesized during the G1 phase. Cyclin D1 forms complexes with CDK4 or CDK6 to drive G1/S transition by inactivating Rb and modulating CDK inhibitors such as p21Cip1 and p27Kip1.
Without growth factor stimulation, Cyclin D1 is unstable and undergoes ubiquitin-mediated degradation, contributing to G1/S arrest. As part of the cyclin family, it exhibits periodic expression patterns essential for cell cycle regulation. Dysregulation of Cyclin D1 through mutation, amplification, or overexpression is linked to tumorigenesis and makes it a valuable target in cancer and cell cycle research.
