Target Information

PAI1 (plasminogen activator inhibitor 1) belongs to the serine protease inhibitor superfamily and is the principal inhibitor of tissue-type and urokinase-type plasminogen activators (tPA and uPA). Platelets are the main source of circulating PAI1, but it is also synthesized and secreted by various cells including fibroblasts, smooth muscle cells, endothelial cells, hepatocytes, and inflammatory cells.
Expression of PAI1 can be regulated by multiple factors such as growth factors, cytokines, hormones, inflammatory factors, glucose or lipid metabolites, and environmental conditions.
PAI1 is elevated in several disease states and has been associated with increased thrombosis risk in obesity, thrombophilia, and metabolic syndrome. Defects in PAI1 can cause abnormal bleeding and are linked to plasminogen activator inhibitor-1 deficiency. PAI1 inhibits fibrinolysis by blocking plasminogen activator activity and may promote neuronal survival. Alternatively spliced transcript variants encoding different isoforms of PAI1 have been identified.
