Target Information
Thymidine kinase-2 (TK2) is a deoxyribonucleoside kinase that phosphorylates thymidine, deoxycytidine, and deoxyuridine, as well as antiviral and anticancer nucleoside analogs. Both recombinant and native forms exhibit broad substrate specificity and complex kinetics, implying a role in activation of chemotherapeutic nucleoside analogs. High expression levels are found in testis and ovary. The main supply of deoxyribonucleotides (dNTPs) for mitochondrial DNA synthesis is derived from the salvage pathway initiated by deoxyguanosine kinase (DGK) and TK2. Mutations in these kinase genes are associated with mitochondrial DNA depletion, suggesting their involvement in maintaining balanced mitochondrial dNTP pools.
