Both high molecular weight DNA (>1 kb, cell-associated) and small DNA (150–250 bp, circulating) are efficiently isolated using a rapid spin column protocol. Salts, proteins, and other contaminants are removed to yield inhibitor-free DNA.

This kit is compatible with Norgen’s Urine Collection and Preservation Tubes.

Background

Urine DNA consists of:

  • Genomic DNA (gDNA): >1 kb, derived from exfoliated cells
  • Apoptotic DNA: 150–250 bp, partly from circulation and may include RNA/DNA hybrids
    Both species can be reliably isolated using this kit.

Sample Storage:
Use Norgen’s Urine Preservative for nucleic acid and protein preservation at ambient temperature.

Centrifugation:
Fixed-speed centrifuge may reduce yields; perform all steps at room temperature.

Processing Variable Volumes:
Protocols can be adjusted for 50 μL–2.6 mL urine by scaling reagent volumes.

Frozen Samples:
Thaw gently at room temperature or 37°C for 5 min; do not centrifuge before processing.
