Target Information

TXK, along with the close relatives BMS, ITK, and BTK, signal downstream of antigen receptors and other types of receptors. All members of this family contain SH3 and SH2 domains and, with the exception of Txk and Dsrc28C, also contain pleckstrin homology (PH) and Tec homology (TH) domains in their amino termini.

Four alternatively spliced forms of Tec are expressed broadly in hematopoietic cells and hepatocytes. The Emt gene product associates with CD28 and becomes activated upon CD28 ligation. Btk is essential for proper B cell development, and mutations in the Btk gene are associated with X-linked agammaglobulinemia (Bruton’s disease). The Bmx protein shows high homology with Btk and is highly expressed in the heart. Txk expression is T cell-specific, while the Drosophila Tec homolog Dsrc28C shows developmentally regulated expression.
