Product Specific Information

A 1:500 dilution of MA5-45935 was sufficient for detection of Tau in 10 µg Baculovirus by dot blot analysis using Goat Anti-Rabbit IgG:HRP as the secondary antibody. Detects approximately 79 kDa.

Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, functioning as a linker protein. Axonal polarity is determined by TAU/MAPT localization in the neuronal cell body domain defined by the centrosome. Short isoforms contribute to cytoskeletal plasticity, while longer isoforms stabilize it. Hyper-phosphorylated Tau forms paired helical filaments (PHF), a major component of neurofibrillary lesions in Alzheimer’s disease. This modification impairs microtubule binding, leading to destabilization and neuronal degeneration. Tau phosphorylation is mediated by kinases such as GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is also found in other CNS disorders including Pick’s disease, frontotemporal dementia, cortico-basal degeneration, and progressive supranuclear palsy.
