Target Information
Members of the TNF superfamily regulate immune responses and induce apoptosis. Two novel members, BAFF (BLyS/TALL-1/THANK/zTNF4) and APRIL (TALL-2), have been identified.
BAFF acts as a B cell activator, inducing proliferation and immunoglobulin secretion, while APRIL regulates both immunological and non-immunological cell growth.
Three receptors—BCMA, TACI, and BAFF-R—bind BAFF and APRIL. TACI, like BCMA, plays a role in regulating humoral immune responses, activating NF-κB and JNK pathways, and is involved in B cell–associated autoimmune diseases.
