Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 패밀리에 속하며, 수용체 관련 키나제에 의해 인산화되어 핵으로 이동 후 전사 인자로 작용합니다. CREB-binding protein 등의 코액티베이터가 STAT3의 전사 활성화에 필요합니다. STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있습니다. Tyrosine 705의 인산화는 JAK1 및 JAK2에 의해 일어나며, 이는 STAT3 이합체 형성, 핵 이동, DNA 결합에 필수적입니다. 인간의 STAT3 유전자는 17번 염색체 q 팔에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. STAT3는 다양한 세포 자극에 대한 유전자 발현을 매개하여 세포 성장과 세포사멸 등 여러 세포 과정에서 핵심적인 역할을 합니다. Rac1 GTPase는 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특정 억제제입니다. STAT3는 세 가지 대체 스플라이싱 변형체를 가지며, STAT3 유전자 결실 생쥐는 초기 배아 단계에서 치명적입니다.
