Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, playing key roles in regulating cell death and stress responses.
TRAF proteins share a conserved region enabling receptor and TRAF interactions, activating distinct signal cascades depending on the TRAF involved.
TRAF2 and TRAF3 directly bind to CD40, a member of the NF receptor superfamily, and are critical for NF-kappa-B activation in mouse B lymphocytes.
TRAF2, together with TRAF6, is required for CD40 signaling in nonhematopoietic cells.
Additionally, TRAF2 interacts with the lymphotoxin-beta receptor (LTbetaR) alongside TRAF3 and apoptosis inhibitors cIAP1 and Smac.
