Target Information
Amyloid beta peptide (Abeta/Beta-amyloid) is the major constituent of amyloid plaques in the brains of individuals afflicted with Alzheimer’s disease. Abeta peptide (40–43 amino acids long) is generated from the beta-amyloid precursor protein (beta APP) through sequential cleavage by beta-secretase (BACE) and gamma-secretase.
Gamma-secretase, a multi-subunit complex containing presenilin-1 and -2, along with nicastrin, modulates Abeta production. Abeta deposits are found in amyloid cores, neuritic plaques, and neurofibrillary tangles, characteristic of Alzheimer’s disease and other neurodegenerative disorders such as Lewy body dementia, Down’s syndrome, amyloidosis, and cerebral amyloid angiopathy.
