이 단일클론 항체는 ZO-1α+ 및 ZO-1α− 아이소폼을 모두 검출합니다. ZO-1 단백질에 특이적이며, 관련 단백질 ZO-2와의 교차 반응은 관찰되지 않았습니다. 인간(Caco-2 세포주) 및 개(MDCK 세포주)에서 반응성이 확인되었으며, 서열 상동성에 따라 다른 종에서도 반응할 가능성이 있습니다.
Alexa Fluor 488의 여기/방출 파장은 각각 495 nm/519 nm입니다. 각 로트는 면역형광법으로 특이성이 검증되며, 권장 보관 조건에서 수령일로부터 1년간 안정합니다.
Target Information
ZO-1은 ZO-2 또는 ZO-3과 복합체를 형성하며, 클라우딘(claudin), 오클루딘(occludin), F-actin과도 결합하여 세포골격과 타이트 결합(tight junction)을 연결합니다. ZO-1 발현은 여러 유방암 세포주에서 현저히 감소하며, ZO-2 및 ZO-3은 상피세포 타이트 결합에서 보편적으로 발현됩니다.
ZO-1은 세포 간 타이트 결합의 세포질막 표면에 위치하며, 세포 간 신호전달에도 관여할 수 있습니다. ZO-1 유전자는 두 가지 아이소폼을 암호화하는 전사 변이체를 가지며, ZO-1 기능 이상은 셀리악병(Celiac Disease) 및 선천성 신증후군(Congenital Nephrotic Syndrome)과 관련이 있습니다.
