Target Information
Hypermethylated in cancer (HIC-1) was originally identified as a target of p53-induced gene expression.
HIC-1 is deleted in the genetic disorder Miller-Dieker syndrome (MDS), and its expression is frequently suppressed in leukemia and various cancers due to hypermethylation and transcriptional silencing.
These findings suggest that HIC-1 functions as a tumor suppressor mediating transcriptional repression.
It is ubiquitously expressed in adult tissues and characterized by five zinc fingers and an N-terminal POZ (BTB) domain.
While other BTB/POZ proteins, such as BCL-6 and PLZF, interact with the SMRT/N-CoR-mSin3A HDAC complex to repress transcription, HIC-1 does not, indicating a distinct repression mechanism independent of the POZ/BTB domain.
