Target Information
DTL/CDT2 acts as a substrate-specific adapter of the DCX (DDB1-CUL4-X-box) E3 ubiquitin-protein ligase complex, essential for cell cycle control, DNA damage response, and translesion DNA synthesis.
The DCX(DTL) complex, also known as CRL4(CDT2), mediates polyubiquitination and degradation of CDT1 and CDKN1A/p21(CIP1), ensuring proper DNA replication regulation.
CDKN1A/p21(CIP1) degradation during S phase or after UV irradiation controls replication licensing.
Substrates interact with PCNA via their PIP-box; those containing the K+4 motif recruit the DCX(DTL) complex for degradation.
In proliferating cells, the DCX(DTL) complex promotes Lys-164 monoubiquitination of PCNA, contributing to PCNA-dependent translesion synthesis.
