Target Information
Monomethyl auristatin F (MMAF) is a synthetic antineoplastic agent. It is part of the approved drug belantamab mafodotin used in multiple myeloma and in experimental antibody-drug conjugates such as vorsetuzumab mafodotin and SGN-CD19A.
In International Nonproprietary Names for MMAF-antibody-conjugates, mafodotin refers to MMAF plus its attachment structure to the antibody.
MMAF is an antimitotic agent that inhibits cell division by blocking tubulin polymerization. When conjugated with an antibody targeting cancer cell structures, MMAF accumulates selectively in those cells.
Chemically, MMAF is desmethyl-auristatin F, meaning the N-terminal amino group has one methyl substituent instead of two as in auristatin F.
