Product Specific Information
This antibody was cross-adsorbed to phospho-tyrosine coupled to agarose and then affinity purified using phospho-IkappaBalpha (Tyr-42) peptide (without carrier).
It detects a 38 kDa protein on SDS-PAGE immunoblots of A431 and Jurkat cells treated with pervanadate, but not in control cells.
Due to homologies to tyrosine sites on other proteins, it is recommended that the antibody be used to detect phosphorylation of immunoprecipitated IkappaBalpha.
Target Information
IkB-alpha is a 40 kDa protein that functions to inhibit NF-κB activity.
The inhibition occurs via protein-protein interaction between IκB proteins and NF-κB dimers in the cytosol.
The interaction of IκB-alpha with NF-κB masks the nuclear localization sequence of NF-κB, preventing its translocation to the nucleus.
Various stimuli such as TNF-α, IL-1β, chemokines, PMA, growth factors, LPS, UV irradiation, viral infection, and other stresses can activate NF-κB by promoting degradation of IκBα.
In humans, the gene is located on the q arm of chromosome 14.
Activation of NF-κB requires phosphorylation of IκB on specific serine residues (Ser32 and Ser36), leading to its degradation.
IKKα (CHUK) interacts with IκBα and specifically phosphorylates it at these sites.
IKKα is critical for NF-κB activation in response to proinflammatory cytokines and is regulated by NIK, a central NF-κB pathway regulator.
The functional IKK complex contains IKKα, IKKβ, and IKKγ subunits, each essential for IκB phosphorylation.
