Product Specific Information

The D7 monoclonal antibody reacts with mouse Sca-1, an 18 kDa member of the Ly-6 family of GPI-linked surface proteins.
D7 recognizes both Ly-6E.1 and Ly-6A.2 molecules expressed by mouse hematopoietic stem cells, myeloid populations, and peripheral T and B cells.
Non-activated splenocytes from Ly-6.2 strains (C57BL, SJL, 129, AKR, etc.) show higher frequency of D7-positive cells compared to Ly-6.1 strains (BALB/c, C3H, NZB, etc.).
Sca-1 expression increases upon activation regardless of Ly-6 haplotype.
Sca-1 serves as a major phenotypic marker for mouse hematopoietic progenitor/stem cell subsets.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse bone marrow cells, ≤0.5 µg/test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume (10⁵–10⁸ cells/test).
Antibody should be titrated for optimal performance.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Stem Cell Antigen-1 (Sca-1) is a member of the lymphocyte antigen 6 (Ly-6) family.
It is expressed on multipotent hematopoietic stem cells (HSC) and used as a marker of HSC in mice.
Sca-1 is used with lineage depletion antibodies to identify murine HSC and is found in fetal liver, adult bone marrow, peripheral blood, and spleen.
It may be involved in regulation of B and T cell activation.
