Description:
The TY25 monoclonal antibody reacts with mouse B7-DC/PD-L2, a member of the B7 family within the Ig superfamily. B7-DC has a predicted molecular weight of approximately 25 kDa and a short cytoplasmic tail (4 amino acids). It is primarily expressed by subsets of dendritic cells, monocytes, and macrophages. B7-DC binds PD-1 but not CD28/CTLA-4, and its interaction with PD-1 can either co-stimulate or suppress T cell proliferation depending on activation state.
TY25 is a valuable tool for studying the role of B7-DC/PD-L2 in antigen-presenting cell (APC) and T cell interactions, as well as for characterizing its expression in mouse immune tissues.
Applications Reported: TY25 antibody is reported for use in flow cytometry.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes and B7-DC transfected cells. Recommended use ≤0.125 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family that binds to PD-1. Although mainly expressed in dendritic cells, PD-L2 expression can be induced in various immune and non-immune cells depending on cytokine environment. IL-4 strongly upregulates PD-L2, with TNF-α and IFN-γ also contributing to expression. PD-L2 has a higher affinity for PD-1 than PD-L1 and functions as an inhibitory immune checkpoint ligand, regulating T cell activation, proliferation, and cytokine secretion (e.g., IFN-γ, IL-10).
Recent studies show PD-L2 can also stimulate T cells and bind a second receptor, RGMb, which is involved in respiratory tolerance and BMP signaling. The human PD-L2 monomer comprises a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
