Product Specific Information
Description:
The UCHT1 monoclonal antibody reacts with human CD3ε, a 20 kDa subunit of the TCR complex. The ε chain, along with γ and δ subunits, is required for proper assembly and surface expression of the TCR complex. CD3 is expressed on thymocytes and all mature T cells. Crosslinking via immobilized UCHT1 triggers intracellular activation and proliferation pathways.
Applications Reported:
Validated for flow cytometric analysis.
Applications Tested:
Pre-diluted and tested on normal human peripheral blood cells. Use at 5 µL (1.0 µg) per 100 µL test volume. Suitable for 10⁵–10⁸ cells/test.
Fluorophore Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm and is compatible with cytometers using a 405 nm violet laser. Verify instrument compatibility before use.
Staining Recommendations:
When using multiple polymer dye-conjugated antibodies (e.g., Brilliant Violet™, Super Bright™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific polymer interactions.
Fixation Compatibility:
BV480 is not compatible with methanol-based fixation.

Target Information
The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits encoded on chromosome 11. These subunits associate with signaling molecules via tyrosine-based motifs. CD3 is expressed on most T lymphocytes and thymocytes but not on B or NK cells. Abnormal CD3 expression is linked to certain immunodeficiencies and autoimmune conditions.
