Target Information
TLR4 is a member of the Toll-like receptor (TLR) family, playing a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities.
TLR4 is most abundantly expressed in placenta and in the myelomonocytic subpopulation of leukocytes. Mammalian cells respond to lipopolysaccharide (LPS) by activating TLR4, which is part of a multi-protein complex containing CD14, LY96, and TLR4. This complex mediates signal transduction events induced by LPS found in most gram-negative bacteria.
TLR4 aids in recognizing pathogen-associated molecular patterns (PAMPs) on infectious agents and mediates cytokine production necessary for effective immunity. Mutations in the TLR4 gene are associated with differences in LPS responsiveness. Several transcript variants have been identified, though their protein-coding potential remains uncertain. TLR4 is expressed by peripheral blood monocytes, a small population of B-cells, and in human placenta. Studies with TLR4-deficient mice indicate that lipopolysaccharide is the main ligand for TLR4, and deficiency increases susceptibility to Gram-negative sepsis.
