Target Information
KLRB1 (Killer cell lectin-like receptor subfamily B, member 1; CD161) is a type II membrane protein with an external C terminus. It is expressed on nearly all NK cells and a subset of CD3⁺ T cells. KLRB1 forms a homodimeric cell surface protein (40–44 kDa) and plays an inhibitory role in NK cell activity. Activation of KLRB1 stimulates acid sphingomyelinase (SMPD1) and increases intracellular ceramide, as well as activates AKT1/PKB and RPS6KA1/RSK1 kinases, leading to T cell proliferation.
KLRB1 acts as a lectin binding to Gal-α(1,3)Gal and N-acetyllactosamine epitopes, and interacts with CLEC2D/LLT1 ligand to inhibit NK cell-mediated cytotoxicity and interferon-γ secretion.
The protein contains extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
