Product Specific Information

The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse bone marrow cells and splenocytes.
Recommended use ≤ 0.6 µg per test (100 µL final volume; 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample (regardless of number of NovaFluors).
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD44 is a 100 kDa type I transmembrane glycoprotein expressed on leukocytes, brain white matter, and epithelial cells of the intestine and breast. It serves as a receptor for hyaluronic acid and mediates cell-cell interactions, adhesion, and migration. Multiple isoforms exist due to alternative splicing, contributing to functional diversity. CD44 expression may correlate with metastatic potential in certain carcinomas. Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus.
