Product Specific Information

Description:
IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells, with high expression in bone marrow myeloid cells and memory T cells. It functions as an adhesion molecule through binding to hyaluronate.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow cells and splenocytes. Recommended ≤0.6 µg/test (100 µL final volume). Optimize antibody titration for best results.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence signatures minimizing spillover and spread.

Target Information

CD44 is a 100 kDa type 1 transmembrane glycoprotein expressed on human leukocytes, brain white matter, and epithelial cells. It acts as a receptor for hyaluronic acid and mediates cell adhesion, migration, and lymphocyte activation. Expression may increase in certain carcinomas, potentially linked to metastasis. Multiple isoforms exist due to alternative splicing, contributing to functional diversity. Associated diseases include superficial keratitis and lichen sclerosus.
