Product Specific Information
Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human MMP-2. Anti-Human MMP-2-specific antibody was purified by affinity chromatography and then biotinylated.
Sandwich ELISA:
To detect Human MMP-2 by sandwich ELISA (using 100 µL/well), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human MMP-2 (500-P307) as a capture antibody, allows detection of at least 2000–4000 pg/mL of Recombinant Human MMP-2.
Western Blot:
To detect Human MMP-2 by Western blot, use at 0.1–0.2 µg/mL. When used with compatible development reagents, the detection limit for Recombinant Human MMP-2 is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information
Matrix metalloproteinases (MMPs) are proteolytic enzymes capable of degrading connective tissue components. MMPs share a common activation mechanism, a conserved amino acid sequence in the metal-binding active site, and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs play key roles in regulating angiogenesis.
MMP2 is synthesized as a 631-amino-acid proenzyme, activated by cleavage of the first 80 amino acids, and contains propeptide, catalytic, and hemopexin domains. MMP2 is involved in tissue remodeling, and mutations in the MMP2 gene have been associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP2 have been identified.
