Target Information
ADRBK1 (GRK2) is a serine/threonine kinase that phosphorylates G protein-coupled receptors (GPCRs). GRK2 (83 kDa) is one of six identified GRK family members and is ubiquitously expressed in mammals. Upon ligand binding and G protein activation, GPCRs are phosphorylated by GRKs, leading to receptor internalization regulated by beta-arrestin-1 and activation of the Ras–Raf–ERK1&2 signaling pathway. GRK2 activity is modulated by phosphorylation (PKC, Src, ERK1&2) and protein interactions. ERK-mediated phosphorylation on serine 670 inactivates GRK2, forming a negative feedback loop.
