Product Specific Information

Specificity:
This antibody binds CD40 and blocks humoral immune responses to T cell-dependent antigens in a dose-dependent manner.

Target Information

CD40 antigen is a single-chain glycoprotein that belongs to the tumor necrosis factor/nerve growth factor superfamily and shows homology to the Hodgkin’s disease-associated antigen CD30. It is expressed on all B cells except plasma cells, and also found on some epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 plays a key role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
The transcription factor AKNA regulates CD40 expression, influencing homotypic cell interactions. Adaptor protein TNFR2 mediates CD40 signal transduction.
CD40-CD40L interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
