Target Information

Piezo1 is a pore-forming subunit of a mechanosensitive non-specific cation channel that generates currents characterized by a linear current-voltage relationship sensitive to ruthenium red and gadolinium. Structurally, Piezo1 contains 36 transmembrane domains and functions as a homotetramer.

Piezo1 plays a key role in epithelial cell adhesion by maintaining integrin activation through R-Ras recruitment to the ER and subsequent stimulation of calpain signaling. In the kidney, Piezo1 may contribute to detection of intraluminal pressure changes and urine flow sensing. It acts as a shear-stress sensor promoting endothelial cell organization and alignment in the direction of blood flow, and is essential for blood vessel formation and vascular structure in both development and adult physiology.

Defects in the PIEZO1 gene are associated with dehydrated hereditary stomatocytosis and lymphedema-related disorders.
