Background
This gene encodes a type II classical cadherin of the cadherin superfamily. Alternative splicing results in multiple transcript variants, with at least one encoding a preproprotein processed into the mature cadherin. These integral membrane proteins mediate calcium-dependent cell-cell adhesion and consist of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small, conserved C-terminal cytoplasmic domain. Type II (atypical) cadherins lack the HAV cell adhesion recognition sequence found in type I cadherins. CAD12 is specifically expressed in the brain and may play a role in neuronal development, particularly during synaptogenesis.
