Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV. SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Suitable for flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells.
• Recommended usage: 5 µL (0.1 µg) per test (100 µL final volume).
• Cell number: 10⁵–10⁸ cells/test (empirical determination recommended).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton technology utilizing nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV. It is mainly expressed in T helper cells, monocytes, macrophages, and dendritic cells. CD4 contributes to T-cell activation via association with the T-cell receptor complex and Lck kinase. It may also mediate neuronal damage in certain immune-related CNS diseases. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and specific brain and gut regions.
