Product Specific Information
Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels by monocytes. CD4 serves as a receptor for HIV. The SK3 clone blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 recognize distinct epitopes.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

- Flow cytometric analysis
Applications Tested:
- Pre-titrated and tested for flow cytometry on normal human peripheral blood cells
- Recommended use: 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
- Add 5 µL per sample before combining with cells.
- For single-color controls: 5 µL buffer per 100 µL cell sample (10³–10⁸ cells).
- NovaFluor conjugates use Phiton™ technology, enabling engineered fluorescent signatures with reduced spillover and spread.

Target Information
CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed on T helper cells, and also found on monocytes, macrophages, and dendritic cells.
Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain regions of brain and gut. CD4 participates in early T-cell activation through association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases.
Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
