• For Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Formalin-fixed tissue staining requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton. It plays a crucial role in determining cell shape, arrangement of transmembrane proteins, and organization of organelles.
Spectrin forms tetramers composed of alpha-beta dimers in a head-to-head arrangement. The alpha-spectrin gene encodes a protein with 22 spectrin repeats involved in dimer formation.
Weaker tetramer interactions compared to non-erythrocytic alpha spectrin may enhance red blood cell membrane elasticity and deformability. Mutations in this gene are associated with hereditary red blood cell disorders such as elliptocytosis type 2, pyropoikilocytosis, and spherocytic hemolytic anemia.
