Target Information

RUSC1, also known as NESCA, shares with the related protein RUSC2 a common domain structure of RUN, leucine zipper, and SH3 domain with over 30% amino acid identity.
RUSC1 acts as an adapter protein binding to the TrkA receptor and is essential for NGF-induced neurite growth in PC12 cells.
It interacts with IKK-gamma, the regulatory subunit of the IKK complex required for NF-kappa-B activation in signaling pathways such as TNF-R and TLR.
RUSC1 also binds to the E3 ubiquitin ligase TRAF6, mediating RUSC1 polyubiquitination.
Overexpression of RUSC1 inhibits TRAF6-mediated polyubiquitination of IKK-gamma, suggesting a regulatory role in IKK-gamma-mediated NF-kappa-B activation.
