Product Specific Information
Description:
Monoclonal antibody 3DS304M recognizes mouse CD304 (Neuropilin-1), a type 1 transmembrane glycoprotein expressed on regulatory T cells (Treg), NKT cells, dendritic cells, stem cells, neurons, endothelial cells, and some tumor cells. Neuropilin-1 functions as a co-receptor for type 3 semaphorins and VEGF, playing a key role in angiogenesis and neuronal development.
In mice, Neuropilin-1 is expressed on thymus-derived natural Treg cells but not peripherally induced Treg. It can transiently appear on recently activated non-regulatory T cells and serves as a marker for recent thymic emigrants in mouse iNKT cells. Neuropilin-1 forms complexes with TGF beta receptors, activating latent TGF beta and enhancing Smad2/3 signaling.
Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Tested by intracellular staining and flow cytometric analysis of mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00). Recommended ≤0.5 µg per test (100 µL final volume). Optimal concentration should be titrated.
Fluorophore Information:
- Excitation: 488–561 nm
- Emission: 607 nm
- Laser Compatibility: Blue, Green, Yellow-Green
- Recommended filter: 610/20 band pass (PE-Texas Red® equivalent).
- Sensitive to photo-induced oxidation — protect from light.
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal fluorescence loss.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
Neuropilin-1 is a 923-amino-acid membrane-bound co-receptor of tyrosine kinase receptors, containing two CUB domains, two F5/8 type C domains, and a MAM domain. It forms heterodimers with Neuropilin-2 and binds plexin (PLXNB1) to form semaphorin receptors.
Primarily expressed in the nervous system, Neuropilin-1 is involved in cardiovascular development, angiogenesis, axon guidance, neuronal circuit formation, cell survival, and organogenesis. It acts as a receptor for semaphorins and VEGF and regulates VEGF-induced angiogenesis.
Neuropilin-1 may be implicated in advanced prostate and breast cancer progression.
