Target Information
ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homolog of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a critical role in ER-based stress signaling.
IRE1 acts as a sensor of unfolded proteins in the ER, initiating the unfolded protein response by splicing XBP1 mRNA into a potent transcriptional activator.
It is essential for cell viability under ER stress conditions that cause unfolded protein accumulation. Depletion of IRE1p or expression of a dominant negative form can lead to apoptosis under such stress conditions.
