This antibody detects endogenous levels of GRK2 only when phosphorylated at Ser29.
Target Information
ADRBK1 (GRK2) is a serine/threonine kinase. Many G protein-coupled receptors (GPCRs) are phosphorylated by G protein-dependent receptor kinases (GRKs). GRK2 (83 kDa), one of six members of this family, is ubiquitously expressed in mammals. Upon ligand binding and G protein interaction, GPCRs (e.g., α2-adrenergic receptor) are phosphorylated by GRKs, leading to receptor internalization regulated by β-arrestin-1 and activation of the Ras–Raf–ERK1/2 signaling pathway. GRK2 activity is tightly controlled by phosphorylation (PKC, Src, ERK1/2) and protein interactions. ERK phosphorylates and inactivates GRK2 at Ser670 in a negative feedback mechanism.
