Target Information
TRAFs are signal transducers for members of the TNFR superfamily, critically involved in activation of MAP3K family members related to NF-kappaB and JNK activation and apoptosis.
TRAF7, a novel 670 amino acid protein, is the human homolog of mouse Traf7 and consists of an N-terminal RING finger domain, central TRAF domain, a coiled-coil motif, and seven WD40 repeats at the C-terminus.
It has E3-ubiquitin ligase activity for self-ubiquitination, specifically interacts with MEKK3, mediates MEKK3-induced AP1 and CHOP activation, and induces apoptosis through Caspase-independent pathways.
TRAF7 also participates in sumoylation and negative regulation of c-Myb, influencing proliferation and differentiation of hematopoietic cells.
