Target Information

Circadian rhythmicity is a fundamental property across diverse organisms, including animals, plants, and fungi. Regulation of endogenous biological clocks occurs via protein-mediated autoregulatory feedback loops. In mammals, genes encoding members of the basic helix-loop-helix (bHLH) PAS (PER-ARNT-SIM) transcription factor family are key regulators of circadian oscillations. Transactivation of CLOCK-induced genes is mediated through E box enhancer elements (CACGTG) upstream of target genes. CLOCK-ARNT3 heterodimers bind to these E box elements to stimulate transcription. The mammalian homolog of Drosophila per is transactivated by CLOCK, and PER, together with TIM (timeless protein), negatively regulates its own transcription by inhibiting the CLOCK-BMAL1 complex.
