Product Specific Information

The IP26 monoclonal antibody reacts with the alpha-beta chain of human TCR, expressed by the majority of peripheral T cells.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for use at 5 µL (0.8 µg) per test (100 µL final volume).
• Cell Range: 10⁵–10⁸ cells/test, determined empirically.
• Note: Co-staining with OKT3 (anti-human CD3) may obstruct staining.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

Buffer Recommendation:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
Use 5 µL per sample for antibody cocktails or single-color controls.

Target Information

The T cell antigen receptor (TCR) consists of an alpha/beta heterodimer associated with CD3 subunits (gamma, delta, eta, zeta) essential for surface expression.
TCR engagement induces T cell activation, differentiation, and cytokine production.
Most peripheral T cells express TCR alpha/beta, while gamma/delta T cells are mainly epithelial.
Defects in TCR expression can halt surface presentation and are linked to autoimmune diseases and tumor antigen recognition.
Superantigen engagement can cause toxic shock syndrome.
Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and lupus patients, suggesting immune compensatory mechanisms.
Human T cells primarily express TCR alpha-beta with CD4 or CD8; a minor population is double negative (DN), which increases in some autoimmune and immunodeficiency disorders.
