Product Specific Information

Description:
The IP26 monoclonal antibody reacts with the alpha beta chain of human TCR. The alpha beta TCR is expressed by the majority of peripheral T cells.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of human peripheral blood cells. Use at 5 µL (0.6 µg) per test (final volume 100 µL). Cell number can range from 10⁵ to 10⁸ cells/test.

Notes:

• Staining with IP26 may be obstructed when OKT3 (anti-human CD3) antibody is used as a co-stain.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining.
  • Add 5 µL per sample to antibody cocktails prior to combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample.
• NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimized spillover and spread.
• Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).
• Internal testing shows NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer associated with invariant CD3 subunits (gamma, delta, eta, and zeta), essential for surface expression.
TCR activation triggers cellular differentiation, proliferation, and cytokine production.
Most T cells express TCR alpha/beta heterodimers, while gamma/delta T cells are mainly found in epithelial tissues.
Defects in TCR expression can impair immune responses and are linked to autoimmune diseases and tumor recognition.
Superantigen engagement of TCRs can induce toxic shock syndrome.
Autoantibodies to TCR V beta segments have been found in autoimmune diseases such as RA and SLE, potentially modulating immune responses.
