Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays an essential role in T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
CD8α is critical for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Binding of MHC-I/peptide complexes to CD8 recruits Lck kinase, leading to cytokine production, motility, and activation of cytotoxic T lymphocytes (CTLs). Activated CTLs are vital for eliminating pathogens and tumor cells. Differentiation of naïve CD8⁺ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
