Target Information

Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway, is essential for normal growth control and may be defective in tumor cells. It supports nutrient preservation under starvation and normal cytosolic component turnover.
Beclin-1, a key regulator of autophagosome formation, is regulated by Ambra1. Ambra1 associates with Beclin-1 via a central region identified by yeast two-hybrid assay.
Null mutations in mice lead to embryonic lethality with severe neural tube defects, autophagy impairment, accumulation of ubiquitinated proteins, unbalanced proliferation, and excessive apoptosis.
Down-regulation of Ambra1 through RNA interference decreases rapamycin- and starvation-induced autophagy. Multiple isoforms of Ambra1 exist.
