Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation level. It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. CD20 expression coincides with surface IgM expression and regulates calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, and its interaction depends on extracellular signals and BCR aggregation. After receptor ligation, CD20 and BCR colocalize briefly before BCR endocytosis, while CD20 remains on the cell surface. CD20 is a key target for antibody-mediated B-cell depletion therapies, as it is stably expressed on malignant B cells and not internalized or shed after treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
