Target Information
Machado-Joseph disease (MJD) is an autosomal dominant neurologic disorder, identical to spinocerebellar ataxia-3. The MJD protein (Ataxin-3) contains (CAG)n repeats in the coding region; expansion from normal 13–36 to 68–79 repeats causes the disease. There is a negative correlation between age of onset and CAG repeat numbers. Ataxin-3 interacts with transcription regulators (CBP, p300, and PCAF), represses transcription, and functions as a histone-binding deubiquitinating enzyme.
