Antibody detects endogenous levels of SYK only when phosphorylated at Tyr525 and Tyr526.
Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases considered potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer.
Syk translocates to the plasma membrane upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, phosphorylating downstream adaptor proteins and initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription.
Syk binds receptor assemblies via its SH2 domains interacting with ITAM motifs phosphorylated by Src-family kinases, which also activate Syk through phosphorylation of its activation loop.
